Determination of biocontrol capacity of endophytic bacteria to the secondary spread of Clavibacter michiganensis subsp. michiganensis in tomato

چکیده
Bitki geli?imini te?vik eden bakterilerin (PGPB) alt grubu olan Endofit bakteriler (EB) bitki dokular? içinde de bulunabilmeleri nedeniyle hastal?klar? ile sava??mda önemli bir potansiyele sahiptir. Bu çal??mada dört farkl? endofit bakteri izolat?n?n domateste Clavibacter michiganensis subsp. (Cmm) patojeninin neden oldu?u bakteriyel kanser ve solgunluk hastal???n?n kontrolu? geli?imi u?zerine etkilerinin belirlenmesi amaçlanm??t?r. Torf/perlit kar???m?nda yeti?tirilen domates fidelerine 108 CFU/mL yo?unlu?undaki EB su?spansiyonlar? kotiledon yaprak a?amas? dördu?ncu? bile?ik olu?um a?amas?nda olmak u?zere iki defa içirme metodu uygulanm??t?r. Cmm’nin su?spansiyonu ise ikinci uygulamas?ndan 48 saat sonra fidelerin u?çu?ncu? yapra??n?n u?stu?ne inokule edilmi?tir. Çal??malar 24±2 oC derecede, %40-60 nem 14 ???k/10 karanl?k ko?ullar?na sahip iklim odas?nda yu?ru?tu?lmu??tu?r. Patojen inokulasyonundan 7 hafta 0-4 skalas? hastal?k ?iddeti, geli?im parametreleri klorofil içeri?i tespit In vitro çal??malarda izolat?ndan u?çu?nu?n patojen s?n?rland?rd??? gözlenmi?tir. izolatlar?n ayn? zamanda in vivo saks? çal??malar?nda da belirlenmi?tir. T2K2-1 izolat? %40 etki en ba?ar?l? izolat olurken, bunu %17,5 15,5 V30G2 T14K1-1 izolatlar? takip etmi?tir. ?zolatlar aras?nda farkl?l?k olsa parametrelerine pozitif katk? sa?lad??? Yaprak say?s?nda, V35Y1 izolatlar?n?n uyguland??? bitkilerde art?? gözlemlenirken bask?s? alt?nda bu gözlenmemi?tir. boyu bak?m?ndan uygulamalar fark belirlenmemi?tir. Kullan?lan EB, biyolojik sava? mekanizmas? s?n?rland?rabilmesi geli?imine ortaya koymalar?,bitkisel u?retimde pestisit sentetik gu?bre kullan?m?n?n azalt?lmas?na yard?mc? olabilecekleri hipotezimizi desteklemi?tir.
